In a randomized, double-blind, placebo-controlled trial, 80 healthy middle-aged adults took NMN for 60 days and blood NAD+ rose significantly in every dose group, with the biggest jump at 600 and 900 mg.[1] A Japanese trial confirmed the same NAD+ rise in healthy older men at 250 mg per day.[2] So "NMN raises NAD+ in people" is genuinely established.

The benefits you would actually feel are a more cautious story. The same 80-person trial found walking distance on a six-minute test improved more in the NMN groups than placebo.[1] A six-week study in 48 amateur runners found NMN improved aerobic capacity markers during exercise, apparently by helping muscle use oxygen more efficiently.[3] And the strongest single result is a carefully controlled trial in which 250 mg of NMN daily improved muscle insulin sensitivity in prediabetic, overweight postmenopausal women.[4] These are real signals — but each is small, short and, in the case of the insulin result, limited to one specific population.

Where the benefits fall short

Honest sourcing means showing the trials that came up empty. A 2025 systematic review and meta-analysis that pooled the randomized NMN and NR trials in older adults concluded the current evidence does not support these supplements for preserving muscle mass, grip strength or gait speed.[5] And a rigorous 12-week trial of the related precursor NR in obese men found no improvement in insulin sensitivity or body composition using the gold-standard clamp method, though it confirmed safety.[6] When you combine the studies statistically, the small individual wins largely wash out. The bottom line on benefits: NMN reliably raises NAD+, shows scattered early hints of physical benefit, and has never been shown in a human trial to slow aging or extend lifespan.
